Mitochondrial toxicity is a major contributor to drug-induced toxicity. Many cells in culture can circumvent mitochondrial insult by increasing ATP production. Mitochondrial dysfunction can remain undetected by the cell viability assay approach measuring ATP. True mitochondrial toxicity would be expected to result in a decrease in oxygen consumption and an increase in extracellular acidification due to generation of lactate through compensation by glycolysis.
